PS: I Remember that had previously written about Yamanaka factors. Rapamycin is a mTOR inhibitor. It was isolated in 1972 from a bacterium found on Easter Island, aka Rapa Nui – hence the name. For many years it was an obscure transplant drug but in the early 2000s was found to significantly extend the lifespan of worms, yeast, flies and mice. It has been widely tested in animals.
The mTOR signaling pathway is also associated with cancer, arthritis, insulin resistance, osteoporosis, and other diseases. Also mTOR signaling pathway is a hot target in anti-tumor therapy research.

TFS. Longevity research is one of my favorite topics on science. David Sinclair, George Church and some other scientists are working on reversing age using Yamanaka Factors with small molecules instead on gene therapy.
Copied...an old comment I made for another post.

"Dr David Sinclair is very well known in this field. The Harvard team has been working for quite some time.
He is the author of "Lifespan: Why We Age―and Why We Don't Have To"
He believes old age is a disease and can be treated one day. His team is essentially looking for the polish to get the cell to read the genome correctly again.
Their work of turning on "Yamanaka Factors" - OSKM led to these developments. They showed that trio of Yamanaka factors (OSK) effectively made cells younger without causing them to lose their identity (i.e., turning back into induced pluripotent stem cells) or fueling tumor growth even after a year of continuous treatment of the entire body of a mouse.

PS: For non-medical general audience there are several books by Dr Nessa Carey which nicely describes the World of Epigenetics. Read them if you can.

The Epigenetics Revolution - How Modern Biology Is Rewriting Our Understanding of Genetics, Disease, and Inheritance
Junk DNA - A Journey Through the Dark Matter of the Genome
Hacking the Code of Life - How gene editing will rewrite our futures"
