Novavax NVX-CoV2373 is a prefusion protein vaccine candidate. It uses Novavax' proprietary recombinant nanoparticle technology with their saponin-based Matrix-M adjuvant technology. Matrix-M is composed of 40 nanometer particles based on saponin extracted from the Quillaja saponaria Molina bark together with cholesterol and phospholipid.
The vaccine is based on an engineered baculovirus containing a gene for a modified SARS-CoV-2 virus spike protein. The baculovirus then infects a culture of Sf9 moth cells, which generate the spike protein and display it on their cell membranes. The spike proteins are harvested and assembled onto a synthetic lipid nanoparticle each displaying up to 14 spike proteins.
Studies with a total of 25,452 volunteers from the US and Mexico. About 48.2% of participants were female, 75.9% were White, 11% were Black/African American, 6.2% were Native American/Alaska Native, and 21.5% identified as Hispanic/Latino. The median age was 47, and about 11.8% of participants were 65 years or older. About 17,312 were administered two doses of the NVX-CoV2373 vaccine, while 8,140 received a placebo.
The results were - It was 93.6% protective against Alpha. 100% effective against SARS-CoV-2 strains that were neither variants of concerns nor variants of interest. Vaccine effectiveness was 92.6% against any variant of concern or variant of interest.
Mild to moderate side effects were observed with vaccinated individuals. The adverse events increased after the second dose. No severe safety concerns were reported after vaccination.
