Based on the OP's question, I think this is more based on the 30DC or the 30 Day Challenge which doesn't require you much capital to start with so practically getting all the domain TLDs will set one back just for getting domains.
Here's my take on the original question based on the 30 Day Challenge.
Get a .com if you are targeting a Global audience more so in the US. As you'd probably be using the Google Keyword Tool assigned to the US. If you are targeting a UK only audience which is huge as well being English, you should go for a .co.uk domain.
There's an option to select UK for demographics in the Google Keyword Tool. I'm sure Market Samurai that's touted by Ed Dale will have the feature to target the demographics as well for keyword searches.

Remember, you are trying to get a site up and rank in your keyword niche based on your research and if the competition's too much on Google (US), try Google (UK) - It may be easier to rank there.
Do note that a .co.uk domain name would set you back 2 years instantly to register the domain name so if budget is your concern, again go back for a 1 year on the .com
Don't analyse too much, get things going and once you've made at least $20 after 30 days get the .co.uk or .net whatever.
