Enveloped viruses does require the fusion of viral and cell membranes to allow release of the viral genome. Mostly, when after virus particles bind to the receptors, they are taken into cells via the cell membranes. This process is pH dependent - change of the pH can act as a catalyst to the process - and this does the trick.
When treated with Chloroquine the pH of the endosome is elevated, which prevents this fusion and blocks viral infection.
Several viruses are known to be inhibited by Chloroquine this way.

You don't take drugs like vitamins.
